The NRCH operates a 2024 Optima AUC instrument with multi-wavelength UV absorbance optics and Rayleigh interference optics. The instrument can operate between 0-40 °C, and measure between 190-800 nm in absorbance mode, and operate between 3-60 krpm. Sample volumes range from 0.11-0.46 ml depending on centerpiece used, and concentrations range between 0.05-10 mg/ml for proteins, a little less for DNA. Accessories include a Genesys S10 UV spectrophotometer, a refractometer, 4-position (up to 60 krpm) and 8-position (up to 50 krpm) rotors, 3-mm and 12 mm centerpieces made from titanium and epon-charcoal, as well as alignment tools, calibration masks, an assortment of sapphire and quartz windows, as well as a computing allocation on the NSF-funded ACCESS infrastructure through grant MCB070038-A25 to support hydrodynamic modeling of the resulting data.
